You'll encounter the Tremortusk during The Broken Sky main story quest. The confrontation takes place shortly after you're tasked with obtaining a cannon to break the Bulwark's wall.

Aloy and Kotallo will arrive at the area where the rebels managed to dominate the Tremortusk. You must destroy the mammoth to obtain the cannon.
