While exploring the world of Dead Cells, you can encounter locked doors with a circular symbol on them - as in the picture. It is not possible to open them in the early runs (after you interact with them nothing particularly interesting happens), however, they can be used in the later parts of the game. This section contains information on what to do to get the doors with the circular symbols to open.

The doors can be opened only after getting the first stem cell, or in other words, after defeating the main boss: The Hand of the King - How to open the doors with the round symbols in Dead Cells? - FAQ - Dead Cells Game Guide

The doors can be opened only after getting the first stem cell, or in other words, after defeating the main boss: The Hand of the King. With the stem cell (Boss Stem Cell), you can get to the tube in the starting location: Prisoners' Quarters and use it to increase the difficulty level (you can learn more about this in the: How to change the difficulty level in Dead Cells? section of this guide).

After activating at least one stem cell, you can start your hunt for the doors. If the symbol on the door is glowing, you can walk through (as in picture 1). If the door is still locked, it means that you do not have the required stem cells (the door in picture two shows a door with a minimum requirement of four stem cells).

Every door leads to a small separate section of the initial level. There, you can encounter new enemies; find new scrolls, new treasure; and meet new merchants. If you encounter a door which is open, you should take your chances and get inside, as the potential loot will give you an advantage in the later stages.
