On this page of the BioShock game guide, you will find the walkthrough of return to Arcadia. Re-visiting this place is intended to create a so-called Lazarus vector and "healing" the garden in order to unlock further passage. Unfortunately, this also means the need for a large battle with Splicers loyal to Ryan.

Before you can build a Lazarus vector, you need 7 more bottles of chlorophyll. Most likely, you accidentally picked them up while exploring Rapture so far. If not, it's best if you go to one of the two Houdini shrines in this level and look there.

Return to the Langford lab and use the U-Invent station to create the Lazarus Vector. Go to Langford's office and use the Central Misting Control.

According to Atlas's advice, run to the lab entrance and use the lever to close it. You have a long battle ahead with Ryan's minions. The best way to succeed is to hypnotize the Big Daddy walking around the neighborhood.
